MICROBIAL BIOSURFACTANT SCREENING: DIVERSITY IN ASSESSMENT METHODS

Abstract: Biosurfactants are a group of surface-active agents mainly produced by microorganisms and have a wide range of applications in industries as well as agriculture. Global usage of such biologically active compounds as detergents, bioremediation agents, cosmetic products is hugely dependent on their high production. New avenues of research have opened up due to this increased interest, most of which are focused on identifying specialized microorganisms capable of biosurfactant production. Screening methods for selection of biosurfactant producing bacteria are, therefore, of utmost importance in this endeavor. The aim of this review was to discuss various conventional and novel methods that can overcome the hurdles encountered in time effective, economical microbial biosurfactant screening. This review lists several important biosurfactant screening methods based on supernatant, biomass as well as culture media. New methods devised for biosurfactant screening in the last half decade such as surfactant dependent dye solubilization, metagenomic studies etc. are also explained in detail. A thorough grasp of both contemporary and anachronistic screening methodologies will not only lead to discoveries of novel native strains capable of biosurfactant production but also form the basis on which new screening methods can be devised

DIAGNOSTIC USEFULNESS OF PCR IN THE RECOGNITION OF LYME DISEASE

Streszczenie: Borelioza jest wieloukładową chorobą wywoływaną przez bakterie zaliczane do kompleksu Borrelia burgdorferi sensu lato. Wektorem przenoszącym infekcję są kleszcze z rodzaju Ixodes, które zakażają kolejnych żywicieli krętka podczas spożywania ich krwi. Zróżnicowany przebieg boreliozy uniemożliwia jej rozpoznanie na podstawie objawów klinicznych. W związku z tym diagnostyka choroby z Lyme opiera się głównie na metodach laboratoryjnych, zarówno bezpośrednich (wykrycie obecności DNA lub białek czynnika zakaźnego w materiale biologicznym pobranym od pacjenta) jak i pośrednich (badania serologiczne). Powszechnie rekomendowanym
podejściem jest serodiagnostyka, jednakże z powodu czasu wymaganego do wytworzenia przez organizm swoistych przeciwciał nie jest ona przydatna w początkowym okresie infekcji. Metody mikrobiologiczne, będące złotym standardem w wykrywaniu wielu infekcji bakteryjnych, nie są szeroko wykorzystywane w rozpoznaniu boreliozy ze względu na wysokie wymagania wzrostowe

od wielu czynników dlatego konieczne jest opracowanie wystandaryzowanego algorytmu diagnostycznego zapewniającego powtarzalność wyników we wszystkich laboratoriach.

Abstract: Lyme disease is a multisystem disease caused by bacteria belonging to the group Borrelia burgdorferi sensu lato. The vector that carries the infection is a tick of the genus Ixodes, that infects subsequent hosts of the spirochete during blood-meal. The varied course of Lyme disease makes it impossible to recognize it on the basis of clinical symptoms. Therefore, the diagnosis of Lyme disease is based mainly on laboratory methods, both direct (detection of the presence of DNA or infectious agent proteins in the biological material collected from the patient) and indirect (mainly serological tests). A commonly recommended approach is serodiagnosis, however, due to the time required for the body to produce specific antibodies, it is not useful in the earliest period of infection. Microbiological diagnostics also can not be used to diagnose Lyme disease in the first weeks of the disease due to its low sensitivity and long waiting time for the result. The solution seems to be molecular diagnostics based on the detection of the spirochete DNA using PCR reaction that is highly specific and sensitive. However, the effectiveness of this approach depends on many factors, therefore it is necessary to develop a standardized protocol ensuring reproducibility of results in all laboratories.

Abstract: The aerobic granular activated sludge process is a promising technology for compact wastewater treatment plants. This system is superior to conventional activated sludge processes in terms of high biomass retention, high conversion capacity, less biomass production, excellent settleabilty and resistance to inhibitory and toxic compounds. The majority of research on granular sludge has focused on optimization of engineering aspects related to reactor operation with little emphasis on fundamental microbiology. The combination of physico-chemical characteristics of granular sludge and the new microbiological methods may provide conceptual information benefiting start-up procedures for full-scale granular-sludge reactors. This article presents a short review of microbiological fingerprinting techniques such as denaturing gradient gel electrophoresis (PCR-DGGE), SDS-PAGE or fluorescence in situ hybridization (FISH) and other techniques such as transmission and scanning electron microscopy (TEM, SEM) or confocal microscopy (CLSM) to give additional information about the structure and bacterial composition of granules.

Abstract: Various species of Candida spp. and Aspergillus spp. remain the most common cause of invasive fungal infections, but other yeasts and filamentous fungi are emerging as significant pathogens. Invasive mycoses continue to be a significant cause of morbidity, especially in transplant recipients and/or in patients undergoing long-time antibiotic therapy. A reliable and early diagnostic method of fungal infections by detecting fungal DNA or invasiveness factors, such as (1→3)-β-D-glucan or antimycelial antibodies, have proven very useful in reducing associated mortality in affected patients. This article is a review of the literature on current serological and molecular methods used in invasive fungemia diagnostics.
